Cornelia Bolen was born on September 11, 1907, the eighth of nine children born to Henry H. and Stella Calhoun Bolen, in Jackson, Alabama. She graduated from the State Secondary Agricultural School in Jackson and finished training as a registered nurse at the Providence Hospital in Mobile, Alabama. Her career led her to practice in various locations in Alabama and Florida, in both hospitals and as a public health nurse in rural areas.

Bolen eventually returned to Jackson where she cared for her aging parents until they passed away. Afterwards she worked in a local nursing home until she retired. She died on January 8, 1972, after a lengthy illness.
